Stem cell factor (SCF) is the ligand of the c-Kit oncogene and is expressed by various structural and inflammatory cells in the airways. Binding of SCF by the c-Kit receptor leads to homodimerization of the receptor and the activation of signalling pathways such as PI-3, PLC-gamma, Jak/STAT, and MAP kinase pathways. SCF expression leads to the induction of mast cell survival and the expression and release of histamine, pro-inflammatory cytokines and chemokines. The inhibition of the SCF/c-Kit pathway leads to a decrease in histamine levels, mast cell and eosinophil infiltration, IL-4 production and airway hyperresponsiveness, suggesting this pathway may be a useful therapeutic target in inflammatory diseases such as asthma. At least two isoforms of SCF are known to exist. Purity: >95% pure by SDS-PAGE analyses. Activity: The biological activity of recombinant human Stem Cell Factor was determined by measuring the proliferation of the factor-dependent human T-helper cell line, TF-1. The ED50 (or 1 unit of activity) for this effect in our laboratories is typically <2ng/ml, corresponding to a specific activity of >5 x10^5 units/mg. Use: For in vitro applications. A dose-response assay should be performed to determine the optimal concentration for specific experiments. Reconstitution and Dilution: Reconstitute lyophilized material in 0.1 ml of 10 mM Acetic acid to a final concentration of 0.1mg/mL. Dilute in a buffer-containing carrier protein, aliquot, and store at or below -70°C. Avoid multiple freeze-thaw cycles. PBS with 1% BSA or a neutral media containing <10% FCS are suitable buffers.
