Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human TSLP. Anti-Human TSLP-specific antibody was purified by affinity chromatography employing an immobilized Human TSLP matrix. Sandwich ELISA: To detect Human TSLP by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human TSLP (500-P258Bt) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant hHuman TSLP. Western Blot: To detect Human TSLP by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant Human TSLP is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

TSLP is a 159 amino acid four helix-bundle hemopoietic cytokine that signals through a heterodimeric receptor complex consisting of the human TSLP receptor and the IL-7R alpha-chain. It impacts myeloid cells and induces the release of T cell-attracting chemokines from monocytes, enhances the maturation of CD11c (+) dendritic cells, naive CD4 (+), and CD8 (+) T cells into proallergic effectors, and primes dendritic cells to produce large amounts of IL-12 after CD40 ligand stimulation. TSLP influences the regulation of the positive selection of regulatory T cells and maintenance of peripheral CD4 (+) T cell homeostasis. It induces allergic inflammation by directly activating mast cells and may be involved in the pathophysiology of inflammatory arthritis. It is expressed in heart, liver and prostate.
