DiscoverX PathHunter™ eXpress TYK2 Activity Assay

The PathHunter™ eXpress TYK2 Activity Assay provides an easy-to-use cell based assay to study small molecule drug candidates targeting CTKs. This kit assesses CTK activation via detection of resulting GHR phosphorylation-induced SH2 domain protein recruitment, mediated by TYK2-JAK2 fusion protein. This product uses EFC technology. The included cells overexpress PK-tagged relevant cytokine receptor and EA-tagged SH2 domain protein alongside an engineered fusion of the cytokine receptor binding domain of JAK2 (Janus Kinase 2) to the catalytic domain from the CTK of interest, TYK2. Upon activation of the cytokine receptor, the CRBD-CTK fusion phosphorylates the receptor, leading to SH2-EA recruitment and production of EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates.
