Maximum efficiency, minimal off-target cleavage, the Cas9 RNP complex can act immediately after it enters the cell and is rapidly cleared from the cell, minimizing the chance for off-target cleavage events when compared to vector-based systems.


GeneArt Platinum Cas9 Nuclease is the recombinant Streptococcus pyogenes Cas9 (wt) protein purified from E. coli that can be used for genome editing with CRISPR technology. Cas9 protein forms a very stable ribonucleoprotein (RNP) complex with the guide RNA (gRNA) component of the CRISPR/Cas9 system.

The Cas9 RNP complex can act immediately once it enters the cell since there are no requirements for transcription or translation as with vector- or mRNA-based systems.

Additionally, the Cas9 RNP complex is rapidly cleared from the cell minimizing the chance of off-target cleavage when compared to other systems (Kim, et al. 2014). Incorporation of a nuclear localization signal (NLS) aids delivery to the nucleus, thus increasing the rate of genomic DNA cleavage.
