Mouse Monoclonal Antibody

The 2.4E6 monoclonal antibody specifically binds to CD212, which is also known as the Interleukin-12 Receptor beta-1 chain (IL12RB1/IL-12RB1/IL-12Rβ1). CD212 serves as a subunit for the functional IL-12 Receptor (IL-12Rβ1-IL-12Rβ2) and IL-23 Receptor (IL-12Rβ1-IL-23R) complexes. CD212 is expressed on T cells, B cells, or NK cells. CD212 is a type I transmembrane glycoprotein that belongs to the hemopoietin receptor superfamily. CD212 is responsible for binding to the IL-12 p40 subunit that is common to the heterodimeric IL-12 and IL-23 cytokines. IL-12 and IL-23 are immunomodulatory cytokines produced mainly by antigen-presenting cells (APC) including monocytes and dendritic cells. These cytokines have overlapping as well as distinct biological activities.

The antibody was conjugated to BD Horizon™ BV421 which is part of the BD Horizon Brilliant™ Violet family of dyes. With an Ex Max of 407nm and Em Max at 421nm, BD Horizon BV421 can be excited by the violet laser and detected in the standard Pacific Blue™ filter set (eg, 450/50nm filter). BD Horizon BV421 conjugates are very bright, often exhibiting a 10 fold improvement in brightness compared to Pacific Blue conjugates. BV421 is a polymer-based dye excited by the violet laser and is one of the brightest fluorochromes offered by BD Biosciences. Conjugates are typically 10 times brighter than Pacific Blue™ conjugates and are often as bright as or brighter than PE conjugates. Due to nearly identical excitation and emission properties but different spillover characteristics, BD Horizon BV421, Pacific Blue™, and BD Horizon™ V450 cannot be used simultaneously.
