Solving the Challenges of Low-level Quantitation in Complex Matrices
The need to improve the ability to identify low-level biomarkers is forcing the analytical community to deal with extremely high levels of analyte and sample complexity, which makes quantitation challenging.
As these proteins are often present at low levels in complex biological matrices, an immunoaffinity capture step is typically employed to purify the sample and increase sensitivity. Following enrichment, the enzyme is activated at elevated temperatures for accelerated digestion under protein denaturing conditions, resulting in a workflow which takes hours rather than overnight.
