MAP3K2 (MEKK2) | |
MAP3K2 was assayed in a two stage coupled reaction involving cascade phosphorylations of inactive MAP2K1 Wild-Type (P3093) and inactive MAPK3 (PV3312) followed by phosphorylation of myelin basic protein (MBP) substrate by activated MAPK3. The enzymes were diluted in enzyme dilution buffer before adding to the reaction. In stage one, MAP3K2 (8.33μg/mL final concentration) is incubated with inactive MAP2K1 (33μg/mL final concentration) and inactive MAPK3 (33 μg/ml final concentration) in 25mM Tris (pH 7.5), 10mM MgCl2, 0.5mM EGTA, 0.5mM Na3VO4, 5mM β-glycerophosphate, 2.5mM DTT, 0.01% Triton™ X-100 and 100 μM ATP for 30 minutes at 30°C. In stage two, 5μL of the above reaction mix is transferred to 25μL of a reaction mix containing 25mM Tris (pH 7.5), 10mM MgCl2, 0.5mM EGTA, 0.5mM Na3VO4, 5mM β-glycerophosphate, 2.5mM DTT, 0.01% Triton™ X-100, 100 μM ATP, 500μg/mL MBP substrate and trace [ 32P]-γ-ATP and incubated for 10 minutes at 30° | |
